Cells cultured with Oxium™Exo secrete higher amounts of exosomes. Our comparative study confirmed the unique benefits that Oxium™Exo provides to exosome-producing cells. First, it maintains cell viability for at least six days without supplementation (FBS or hPL). Second, these cells had significantly higher viability and presented fewer apoptotic and cell death levels than those cultured in DMEM or commercial medium. Third, cells cultured in Oxium™Exo maintain the characteristics of Mesenchymal Stromal Cells (MSCs), as defined by the International Society for Cell and Gene Therapy (ISCT). Fourth, cells cultured in our media produced more than nine times more small extracellular vesicles than competitors after six days. This edge over other media is even greater for those vesicles between 50 to 200 nanometers.

Oxium™Exo-derived vesicles are internalized by target cells and maintain their biodistribution profile. It is critical that a cell culture media that enhances the secretion rate of sEVs also does not modify their structural, biochemical, and classical properties. This positive impact was determined mainly by the internalization capacity of Oxium™Exo-produced sEVs in cartilage and cancer cells in vitro and by the biodistribution profile in vivo. Our research concluded that the surface markers, cell uptake, and biodistribution pattern of sEV are not affected by the use of Oxium™Exo.
